Welcome to the DNA Sample Prep Forum!

Hello and welcome to the DNA Sample Prep Forum! I’m Jill Herschleb and I lead the Sample Prep group at 10x Genomics. We develop protocols and advice for HMW DNA extraction that deliver the optimal performance with the Chromium Genome Solution. These protocols linked in the “resources” section above and also available on support.10xgenomics.com.

We hope that you will use this space to share protocols with each other, ask questions, and brainstorm new solutions to your challenges. We’ll be here to help, to answer your questions, and to share all of our expertise.

When discussing DNA sample prep and QC, it is useful to report the DNA size measured both before and after running Chromium Genome, as there will be some physical shearing that occurs during the assay (this is true for all long-range genomics systems).  For instance, our Demonstrated Protocol for Whole Blood (https://support.10xgenomics.com/genome-exome/sample-prep/doc/demonstrated-protocol-hmw-dna-extractio...) produces DNA ~250 kb on a pulsed-field gel, and ~90 kb after sequencing.

Thanks for stopping by!  -Jill
