During cDNA amplification, why is the elongation time only 1 minute?
One minute might be long enough for the majority of mature mRNAs, but it wouldn't be long enough to amplify a lot of pre-mRNAs, right? This would be especially problematic when working with nuclei instead of whole cells as you'd probably fail to amplify a large proportion of your template molecules. Right?
