Multiplexing using lipid-modified oligonucleotides

Hi all,

 

I was just wondering what thoughts people might have on this technique, which I just came across: 

MULTI-seq: Scalable sample multiplexing for single-cell RNA sequencing using lipid-tagged indices 

 

For those working with nuclei instead of whole cells, this technique seems like the best option for multiplexing.

Hello,

I am curious if you have explored using this technique for nuclei multiplexing?

Additonally, does anyone knows the status of these techniques becoming commercially available?

Raleigh,

 

I still haven't tried any multiplexing yet, but I have my eye on a few other techniques as well:

- Nuclei multiplexing with barcoded antibodies for single-nucleus genomics

which uses antibodies targeting the nuclear pore complex

- Multiplexed droplet single-cell RNA-sequencing using natural genetic variation.
which works if your samples are from different patients (or have any significant degree of genetic differences, I guess)

Cell Hashing with barcoded antibodies enables multiplexing and doublet detection for single cell gen...

if you're working with whole cells instead of nuclei.

 

I don't know of any of these lipid- or anitbody- conjugated antibodies being commercially available, but I haven't looked super hard.
